First, to ignore someone, visit their profile by clicking on their username, then look for the "Ignore" button near the top of the page. You'll be given the option to either "Block Mail" (only blocks mail from the user) or "Full Ignore" (essentially makes the user disappear from your view). Either way, the user will NOT be informed.

Second, you can manage your ignore list by clicking the My DU tab, then "Ignore List" -- from here you can add or remove people from your list.
